Nanodisc consists of an amphipathic protein called membrane scaffold proteins (MSPs), which is embellished versions of the major ingredient of HDL, apolipoprotein A1 (apoA1). Nanodisc is self-assembled soluble discoidal phospholipids bilayers, thus a functional stabilized membrane disk can be supplied by Nanodisc for the combination of membrane associated and membrane-bound molecules. Membrane associated macromolecules are extremely crucial drug targets related to drug metabolism and signal transduction. Nanodisc has been used to solubilize and functionally stabilize a number of membrane proteins, it can offer homogeneous discs with steerable stoichiometry of the incorporated protein.

Nanodisc offers passageways to both sides of the bilayer structure, avoiding the problem of the orientation of the inset membrane protein as can be the case in vesicles and supported membrane mono- or bilayers. What’s more, the ability of modifying the MSP sequence and attaching various tags at particularly engineered sites provides chances for the immobilization of membrane bilayers and membrane-associated molecules, which is available for determination on protein chips and affinity chromatography media.

Creative Biostructure has created two systems to testify the immobilization of lipids and binding of protein analytes. In the first system, Nanodisc contains biotinylated lipids and the delivery of biotinylated lipids are measured by streptavidin labeled quantum dots. Biotin–streptavidin offers a highly firm and well-characterized protein-target binding system which is perfect for demonstrating the feasibility of this technique. However, in the other system Nanodiscs contain a variety of phosphoserine lipids. These Nanodiscs are filtrated with annexin, a family of proteins which act as a crucial part in various cellular functions. Annexin assays are broadly used to estimate apoptosis, which is an essential biological process but not works in several diseases such as cancer and Alzheimer’s.
