Mempro™ Pre-assembled Nanodiscs

Creative Biostructure has made great efforts on developing a magic nanodisc platform in the past years, making us in the leading place to provide a variety of nanodisc products and services. The major products based on our Mempro™ Nanodisc technology are the custom pre-assembled nanodiscs composed of different membrane scaffold proteins (MSP1D1, MSP1E3D1 and MSP2N2) and phospholipids (POPC, DMPC and DMPG).

The most important factors for the optimal assembly of membrane proteins into nanodiscs include the lipid-protein stoichiometry and the careful choice of detergent and temperature. In order to achieve homogeneous sized nanodiscs, it is crucial that an optimal MSP : phospho- lipid ratio is used. If the ratio is far from the optimum, an array of different sized lipoprotein particles are formed alongside with some nanodiscs. Creative Biostructure has attempted to form custom nanodiscs with a new lipid composition and incorporate membrane proteins into nanodiscs by empirically adjusting the ratio of the components to achieve a maximum yield of nanodiscs. The most widely used detergent used to assemble nanodiscs is sodium cholate. Sodium cholate is usually present in the buffer used for assembly at a mole ratio of 2:1 to the total phospholipid amount. This ensures proper solubilization of both MSP molecules and phospholipids and the avoidance of any unwanted precipitation.

Please note that our Mempro™ Pre-assembled Nanodiscs are only for the usage of cell-free expression reactions. We do not recommend you to use them to reconstitute solubilized membrane proteins. DMPC is used as eukaryotic phospholipid while DMPG is used as prokaryotic phospholipid, and biotinylated modifications for Mempro™ Pre-assembled Nanodiscs are also available.
