Migrasome Services
Organelles are the basic structures in a cell, and the different organelles perform their respective functions within the cell to maintain normal cellular activity. Migrasome is a widespread organelle in all types of migrating cells and is found in all types of migrating cells in humans, mice, and rats. The Migrasome helps dorsal forerunner cells (DFCs) to find their place, ensuring the normal formation of Kush vesicles and thus the normal development of organ morphology. The Migrasome is responsible for the maintenance of cellular homeostasis, the integration of intercellular signals, and the transfer of substances between cells.

Migrasome Isolation and Purification
We can isolate and purify migrasomes by differential centrifugation combined with density gradient centrifugation, negative staining by electron microscopy and ultrathin sectioning by electron microscopy, and mass spectrometry of the purified migrasome to obtain information on the protein composition of the migrasome.

Migrasome Assay Marker Identification
Our strategy for identifying migrasome-specific protein markers for our clients is to select candidate proteins based on quantitative mass spectrometry analysis of purified migrasome, followed by validation using cell biology and biochemistry methods.
The migrasome is first isolated and purified (which can be done by the methods described above), labeled using tandem mass tags (TMT), followed by quantitative mass spectrometry to identify proteins enriched on the migrasome, and then we combine and analyze the samples to determine the protein enrichment on the migrasome compared to the cell body. Based on the MS data, proteins that are enriched only on the migrasome can be screened. We can also stain the cells with antibodies specific to these proteins to determine that they are all located in the migrasome.
In addition to this, we can use mass spectrometry with green fluorescent protein (GFP) labeling to identify multiple proteins enriched on the migrasome.
Applications
- Promoting angiogenesis studies. Migrating monocytes act as forerunners to promote angiogenesis by depositing migrasomes enriched with pro-angiogenic factors, creating a favorable microenvironment for angiogenesis.
- Mediating mitochondrial quality control. Both mitochondrial stimulations as well as cellular starvation can significantly increase the number of mitochondria within the migrasome.
