How Photo-activated Liposomes Work

Liposomes constituted from light-sensitive lipids have been explored since early 1980s. Membrane permeabilization of these light-controlled liposomes are induced upon light exposure of specific wavelength to target drug release. A successful example of photo-activated liposome formulation in clinical use now is Visudyne therapy to treat age-related macular degeneration of (AMD) in patients. In this formulation, essential drug release is followed by non-photothermal treatment with a 689 nm laser source after injection. As a candidate for sustained and on demand drug delivery, photo-activated liposomes gained more attention with its various parameters, such as exposure time, wavelength, beam diameter, and laser intensity to adapt to different purposes.   • Light-induced Oxidation Liposomes: Use reactive oxygen species to light-induce oxidization of unsaturated lipids, then induce formation of pores in membrane bilayers.
  • Photocleavable Liposomes: Design susceptible-to-light amphiphiles moiety (typically in the UV range) to generate breakdown products that can destabilize liposome membrane.
  • Photopolymerizable Liposomes: Design photopolymerizable phospholipid molecules that photo-crosslinking under light induce and rendering the unstable bilayers.
  • Photoisomerization Liposomes: Design photoisomerization moiety, the conformational change destabilizes the lipid membrane and allows for release.
  • Molecular Dyes Liposomes: Using hydrophilic dye or hydrophobic dye to achieve cargo release under a wide array of light wavelengths.
  • Gold Nanoparticles Liposome: Development of single nanometer size gold to mediate photothermal conversion in liposomes.
