Tangential Flow Filtration (TFF), also called cross flow filtration, is a powerful and rapid membrane filtration technique for biological macromolecules-protein, virus-protein, cell-protein, protein-salt separation and purification. It can be widely applied in various biological researches such as recombinant protein production, immunology, microbiology, biochemistry, etc. TFF performs the separation based on the different molecular size of biology molecules. TFF differs from the Direct Flow Filtration (DFF). In a TFF assay, samples are forced to run parallel to the TFF filter in a cycle, components with smaller size than the pores of TFF medium membranes will be pushed out through the pores. But components with larger size than the pores will have to flow parallel to the filter in the cycle. By this way, the separation is done according to the molecular size. A reservoir is usually used to improve the TFF assay so that the sample can be recycled through numerous times. TFF is also a wonderful technique for diafiltration and desalting. Various medium filters with different pore size have been developed to meet different experimental requirements.

The TFF process is simple and can be performed easily, TFF experimental system is very easy to set up and handle. With TFF, researchers can perform the higher concentrations with less time than dialysis. What's more, sample concentration and diafiltration can be performed on the same TFF system, saving the time and prevent the losses of sample. It is convenient for TFF to scaled-up or scaled-down, TFF can be applied to concentrate and desalt samples from 10ml to hundreds or even thousand liters with different TFF devices. The cost of a TFF assay is also low, as the devices and cassettes can be reused. The application of TFF is universal and can be involved in multiple experiments such as:
- Concentrate and desalt bio-macromolecules such as proteins, peptides, DNAs, RNAs, etc.
- Fractionate dilute protein mixtures
- Harvest cells.
- Deliver, recover and purify of certain macromolecules from cultured cells, blood or other sources.
- Prepare sample for chromatography.
- Recover or remove viruses.
- Clarify tissue homogenates or cell lysates.
