Mempro™ Detergent-Free Bcl-2 Inhibitors of Programmed Cell Death Production
Creative Biostructure can provide advanced custom Mempro™ B-cell lymphoma 2 (Bcl-2) inhibitors of programmed cell death superfamily protein production services using detergent-free expression system.
Bcl-2 is known as a regulator of apoptosis (programmed cell death), which is encoded in human by BCL2 gene. Bcl-2 is an important anti-apoptotic protein and is therefore known as an oncogene. Bcl-2 family can be divided into two classes of proteins, including pro-death proteins and pro-survival proteins. The pro-death proteins include BCL-2-associated X protein (BAX), BCL-2 antagonist/killer 1 (BAK), BCL-2-associated agonist of cell death (BAD), BCL-2-like 11 (BIM), NOXA, and BCL-2 binding component 3 (PUMA), whereas the pro-survival proteins include BCL-2, BCL-XL, BCL-2-like 2 (BCL-w), myeloid cell leukemia sequence 1 (MCL-1), and BCL-2-related protein A1 (BFL-1). Conventional techniques for transmembrane protein production require extraction from the membrane into detergent micelles. However, the significant drawback of detergent is that can induce conformational changes on the proteins. Our alternative detergent-free expression system can best retain the native conformation of Bcl-2 inhibitor superfamily proteins for further functional analysis.

Creative Biostructure has extensive experience in high-yield Bcl-2 inhibitor superfamily protein production in detergent-free protein expression system, we can employ various strategies for Mempro™ detergent-free Bcl-2 inhibitor superfamily production, including:
- Mempro™ Bcl-2 inhibitor superfamily production using nanodiscs
Nanodiscs are the self-assembled nanotechnology to stabilize transmembrane proteins removed from the membrane by membrane scaffold proteins (MSPs) without detergent. Creative Biostructure can reconstitute Bcl-2 inhibitor superfamily into nanodiscs in detergent-free procedure: Expressing the Bcl-2 inhibitors in the cell-free expression system with the addition of pre-assembled nanodiscs.
- Mempro™ Bcl-2 inhibitor superfamily production using amphipols
The amphipathic amphipols have the ability to enwrap transmembrane proteins, allowing them to stay folded. Creative Biostructure can apply A8-35 to solubilize Bcl-2 inhibitors during purification and stabilization processes.
- Mempro™ Bcl-2 inhibitor superfamily production using poly (styrene-co-maleic acid) lipid particles (SMALPs)
The SMALPs can be self-assembled by the simple addition of the SMA co-polymers. At neutral or alkaline pH, The disc-like structure assembles itself, encapsulating Bcl-2 inhibitors in a formation amenable to be purified.
These novel detergent-free techniques for membrane protein production can be obtained easily, and enabling more comprehensively structural and functional studies.
