Brief Introduction to Isothermal Titration Calorimetry (ITC)
Both intramolecular and intermolecular biochemical reactions involve a reversible heat-driven process, that is, a certain degree of heat change occurs before and after the reaction. The ITC technology utilizes the principle of power compensation, which can accurately obtain the intramolecular/intermolecular interactions of the system in a wide concentration range by single-concentration scanning, directly measure the heat of the reaction, and complete the fitting calculation of combined thermodynamic parameters. By fitting the interaction mode of the two reactants with the software, it is possible to directly calculate the reaction binding enthalpy (ΔH), the number of binding sites, binding equilibrium constant (Ka), constant pressure heat capacity (ΔCp), as well as kinetic data between two or more molecules in the solution. Thus, we can derive the Gibbs free energy (ΔG) and entropy change (ΔS) of the reaction.
