Lipid Nanoparticle (LNP)

Lipid nanoparticle is a new drug delivery system that can dissolve, wrap, adsorb, and attach drugs or other bioactive substances by using biocompatible lipid materials as a carrier. Compared with other drug delivery systems, lipid nanoparticles can improve drug absorption, improve drug stability, change drug toxicity, and have sustained and controlled releasing effects. Therefore, lipid nanoparticle has been widely used as a carrier system for gene drugs, antitumor drugs, proteins, and peptides. Meanwhile, complex and unstable target molecules can also be inserted into the lipid nanoparticles for structural analysis by cryogenic electron microscopy (cryo-EM). So far, lipid nanoparticles have provided effective support for the function and pharmacology of small molecules.

Cryo-EM single-particle analysis technology has become a new force among many structural analysis methods in structural biology, and it is playing an increasingly important role in the structural analysis of mixed lipid nanoparticles. Basic information about the nature and source of particulate matter can usually be gleaned from the size, shape and/or surface form of individual particles.

LNPs have become the golden standard in the nonviral delivery of genes. Our EM platform develops a particle morphology characterization system that can analyze the shape and size of individual particles ranging from 40nm to 2,000 nm. We can also help distinguish individual particles or polymers, quantify structures of polymers, analyze spheres or non-spheres, and count individual particles in mixtures.
